That is, have you ever used it as a giant stompbox? I did it once on a Triton Extreme a long time ago to play rhythm guitar on a cover of Green Day's version of the "Simpsons" theme at a wedding gig (yes, it was a special request). Is it easy to do on a Nautilus?

It shouldn't be too difficult. There's a video of it being done on the Kronos. The Nautilus will be very similar.

https://www.youtube.com/watch?v=9n7ctQ8qgA8

The guy in the video doesn't really give much detail, but the key things to do after connecting the guitar, are to select the input that your guitar is connected to, and route the input into the effects (IFX1)(see 4m37s into the video), and get the levels right, using the controls at the back of the Nautilus. The Programs used in the video utilise the HD-1 engine, and the way to link the inputs to the effects is shown e.g. around 6:23 in the video, though the other sound engines can also be used.

If you've done this on the Triton Extreme, then you probably know this already, but it might be useful for anyone else who might be interested. The effects need to be chained, like a series of pedals are chained together, and this is done in the effects menu pages. See around 6:33 in the video. In the Insert FX tab, check the Chain boxes to do this, and ensure the selected effects are On (the little onscreen buttons next to the IFX1, IFX2, IFX3 labels). The rest should hopefully be a little easier to do and experiment with to get the desired sound.
